Internal standard selection

Hello Iolite community!
I am doing some quantification of trace elements in calcite matrix. If I collect 44Ca signal to be used as an internal standard, should I still input a Ca concentration of 40.04% similar to Paul et al, (10.1039/C2JA10383D ), even if the collected signal is for 44Ca? Also, If I am collected 51V signal, will the output of data reduction be the concentration of vanadium in calcite, or will be the concentration of the collected isotope only?
