This happens when iolite cannot find your DRS folder and/or your python site packages.
We are working on making this more straightforward but the short answer is to follow the instructions in this post (starting where Joe writes "Alternatively...")

The longer answer is that we have perhaps allowed too much flexibility in where the user can store certain important files (such as the 3D Trace Elements DRS file). In future versions of iolite, important files such as this will be in a fixed location, and the user will be able to make modifications to a copy stored elsewhere if they want to.
